Helperby Therapeutics announces phase 1 results demonstrating multiple benefits of its unique combination antibiotic therapy - effective against the WHO Critical Priority Carbapenem resistant bacterial pathogens
Helperby Therapeutics Ltd (Helperby) has presented data at the European Congress of Clinical Microbiology and Infectious Diseases (ECCMID) announcing Phase I results on the tolerability, pharmacokinetics, pharmacodynamics and efficacy of AZT and colistin both alone and as a combined therapy, against multi-drug resistant Enterobacteriaceae (CRE), one of the most serious drug resistant pathogens.

Helperby also presented preclinical data which shows that AZT is active against carbapenem and colistin resistant Enterobacteriaceae. AZT is a DNA chain terminator and represents a new class of antibacterial agent against the most resistant Gram-negative bacteria. Further data were presented showing the combination to be active in animal models.
This research paves the way for further clinical trials and the potential for the development of a new class of antibiotics, effective against some of the most resistant pathogens. This combination could reach the market in just three to five years.
The significance of these data is that the WHO Critical Priority Gram-negative Carbapenem Resistant bacterial pathogens will contribute to the 10 million deaths which are predicted worldwide by 2050 from treatment-resistant infectionsi. The increasing resistance of pathogens to antibiotics is a serious threat to modern medicine. Blood-stream infections with these highly resistant bacteria can result in fatality rates of over 50%ii.
The combination provides antibiotic enhancement with a low dose of colistin, which is safe and tolerable in humans as demonstrated by the phase 1 results, and so avoids the risk of renal toxicity associated with colistin in higher doses.
Helperby’s new class of ARB antibiotic therapy is one of only a handful of drug candidates that the World Health Organisationiii has identified as in development to tackle the three WHO Critical Priority pathogens; Carbapenem-resistant Enterobacteriaceae (CRE), carbapenem-resistant Acinetobacter (CRAB) and carbapenem-resistant pseudomonas aeruginosa(CRPA).
AZT and colistin are known entities and their mode of action, side-effects and interactions are well known by regulatory authorities.
“Our unique approach of combining antimicrobial resistance breakers such as azidothymidine with existing antibiotics has the potential to reach the market with a new class of effective antibiotic therapy in 3 to 5 years”, said Professor Coates, founder and Chief Scientific Officer at Helperby Therapeutics.
“Helperby has developed a strong candidate for a longer lasting therapy effective against multiple drug- resistant infections,” says CEO Dennis Molnar. “It’s well established that combination therapies prevent the emergence of resistance, and Helperby’s combination technique has the potential to extend the life of last-resort antibiotics which are under huge threat from the accelerating emergence of antimicrobial resistance.”

Janssen Phase 3 Study Programme of Esketamine Nasal Spray in Patients with Treatment-Resistant Depression Presented for the First Time in Europe19.6.2018 09:00 | Tiedote
The Janssen Pharmaceutical Companies of Johnson & Johnson presented for the first time in Europe data from pivotal Phase 3 clinical studies of the investigational compound esketamine nasal spray in treatment-resistant depression.1,2,3,4 The studies, conducted by Janssen Research & Development, LLC, were presented at the 31st International College of Neuropsychopharmacology (CINP) congress in Vienna, Austria. Data discussed from the Phase 3 programme included results from the maintenance phase of a long-term relapse prevention study in adults with treatment-resistant depression. The data found that continuing treatment with esketamine nasal spray plus an oral antidepressant in patients beyond 16 weeks showed clinically meaningful and statistically significant superiority to treatment with an oral antidepressant plus placebo nasal spray in delaying time to relapse of symptoms of depression. Furthermore, the data indicated that patients in stable remission treated with esketamine nasal sp

Inside Secure to Provide Secure Provisioning Solutions to NationalChip18.6.2018 18:35 | Tiedote
Regulatory News: Inside Secure (Paris:INSD), at the heart of security solutions for mobile and connected devices, announced it entered into an agreement with Hangzhou NationalChip Science and Technology Co., Ltd, a leading innovator and developer of SoCs (system-on-chip), to use Inside Secure’s Secure Provisioning Solution to build robust security into its products during the chip manufacturing process. Secure provisioning is recognized as a crucial, foundational security building block across diverse markets including automotive, entertainment, IoT, mobile and TV. Securing devices or end products can be worthless if their identities and cryptographic keys have been compromised during manufacturing. Widely-publicized attacks have targeted provisioning because they can affect large numbers of devices and are difficult to detect. Chipmakers and device manufacturers see the potential for new revenue sources by offering secure provisioning to their customers, leveraging technology solution
